Basic concepts. Stable isotopes of carbon and oxygen. Fractionation, cases and effects.Stable carbon isotope: δ13C in the atmosphere, δ13C in tree rings. Physiological processes (diffusion through the stoma, Rubisco activity, concentration of δ13C in sugars). Environmental signal. Efficiency in water use (calculation andinterpretation). Juvenile effect.Stable isotope of δ18O oxygen: δ18O in the precipitation, soil and water of the xylem. δ18O in tree rings. Evaporation atleaf level (fractionation, Péclet Effect). Fractionation during the formation of sugars. Isotopic exchange with xylemwater. Concentration of δ18O in sugars. Environmental signal. Practical and methodological aspects. Sample preparation. Sampling in the field. Criteria for the selection of the sitefor dendro‐isotopic studies. Adequate number of trees and samples. Preparation and cutting of growth rings (wholerings, annual samples or pooling), milling. Methods of chemical extraction from wood to cellulose homogenization.Cellulose homogenization. Spectrometric measurement (replicas, reference materials, etc.).How to make long chronologies (cohorts, superimposed periods, etc.). Analysis of data from δ13C in tree rings.Calibration of raw data from δ13C of the spectrometer using reference materials.
